Cloning and functional characterization of seed-specific LEC1A promoter from peanut (Arachis hypogaea L.)

LEAFY COTYLEDON1 (LEC1) is a HAP3 subunit of CCAAT-binding transcription factor, which controls several aspects embryo and postembryo development, including morphogenesis, storage reserve accumulation skotomorphogenesis. Herein, using the method chromosomal walking, 2707bp upstream sequence from ATG initiation codon site AhLEC1A homolog Arabidopsis LEC1 was isolated in peanut. Its transcriptional start confirmed by 5’ RACE located at 82 nt ATG. The bioinformatics analysis revealed that there existed many tissue-specific elements light responsive motifs its promoter. To identify functional region promoter, seven plant expression vectors expressing GUS (?-glucuronidase) gene, driven terminal series deleted fragments were constructed transformed into Arabidopsis. Results histochemical staining showed regulatory containing 82bp UTR 2228bp promoter could facilitate to express preferentially embryos different development periods Taken together, it inferred during seed peanut might be controlled positively seed-specific elements, as well negatively some other inhibiting organs. Moreover, pattern transgenic seedlings darkness relevant light-responsive scattered segment, implying these harbored regulate skotomorphogenesis seeds, inhibited after germinated transferred light.
